چکیده

The gestational state is a period of particular vulnerability to diseases that affect maternal and fetal health. Stress during gestation may represent a powerful influence on maternal mental health and offspring brain plasticity and development. Here we show that the fetal transcriptome, through microRNA (miRNA) regulation, responds to prenatal stress in association with epigenetic signatures of psychiatric and neurological diseases. Pregnant Long-Evans rats were assigned to stress from gestational days 12 to 18 while others served as handled controls. Gestational stress in the dam disrupted parturient maternal behaviour and was accompanied by characteristic brain miRNA profiles in the mother and her offspring, and altered transcriptomic brain profiles in the offspring. In the offspring brains, prenatal stress upregulated miR-103, which is involved in brain pathologies, and downregulated its potential gene target Ptplb. Prenatal stress downregulated miR-145, a marker of multiple sclerosis in humans. Prenatal stress also upregulated miR-323 and miR-98, which may alter inflammatory responses in the brain. Furthermore, prenatal stress upregulated miR-219, which targets the gene Dazap1. Both miR-219 and Dazap1 are putative markers of schizophrenia and bipolar affective disorder in humans. Offspring transcriptomic changes included genes related to development, axonal guidance and neuropathology. These findings indicate that prenatal stress modifies epigenetic signatures linked to disease during critical periods of fetal brain development. These observations provide a new mechanistic association between environmental and genetic risk factors in psychiatric and neurological disease.

Effect of maternal variable stress on oxidative status and glucose metabolism in pubertal male rats

Introduction: Metabolic disorders are affected by negative stress experiences in the early stages of life. Accordingly, in this study, the effects of stress during pregnancy on oxidative status and glucose homeostasis in pubertal male Wistar rats were investigated. Materials and Methods: After pregnancy, female rats (200±30 g) were divided into 2 groups (6 rats per group) of stress and non-stre...

Effect of Predatory Stress Before Fertilization on Body Weight and Pentylenetetrazole-Induced Seizure in Rat Offspring

Background and Objective: Spermatogenesis and oogenesis stress is a primary epigenetic element that can cause lifelong modifications in neural system functions. This study was designed to elucidate whether preconceptional stress during spermatogenesis and oogenesis affects the seizure behaviors and body weight of the offspring later in life. Materials and Methods: In this experimental study, 1...

More than genes: the advanced fetal programming hypothesis.

Many lines of data, initial epidemiologic studies as well as subsequent extensive experimental studies, indicate that early-life events play a powerful role in influencing later suceptibility to certain chronic diseases. Such events might be over- or undernutrition, exposure to environmental toxins, but also changes in hormones, in particular stress hormones. Typically, those events are trigger...

The Effect of Maternal High-Fat Feeding on Energy Homeostasis in Stressed Adult Male Rat Offspring

Introduction: In the present study the effect of chronic maternal high-fat diet consumption on energy homeostasis and glucose metabolism in response to chronic stress was investigated in adult male rats. Materials and Methods: Female rats were divided into two groups of normal and high fat diets. Each group received their diet from 3 weeks before pregnancy until the end of lactation. At 8 weeks...

Impact of Maternal Stress in Pregnancy on Brain Function of the Offspring.

Epidemiological studies suggest that exposure to prenatal stressors, including malnutrition, maternal immune activation (MIA), and adverse life events, is associated with increased risks of schizophrenia, autism spectrum disorder (ASD), and attention-deficit hyperactivity disorder (ADHD). However, the underlying pathophysiological mechanisms are unclear. The first trimester of pregnancy is part...
